In this issue, Mishra and colleagues identify lysine methyltransferases and demethylases that control site-specific DNA re-replication and copy gains. The labeled envelopes in the image illustrate the interplay between lysine-modifying enzymes in modulating the epigenetic "addressing system" for site-specific DNA amplification.
