In this issue, Mishra and colleagues identify lysine methyltransferases and demethylases that control site-specific DNA re-replication and copy gains. The labeled envelopes in the image illustrate the interplay between lysine-modifying enzymes in modulating the epigenetic "addressing system" for site-specific DNA amplification. For example KMT-KDM balance controls the site of DNA amplification (zip code), while the KMT2-KDM4 enzyme network promotes copy gains (sender) and KDM5 blocks such amplifications (postmark).
